Der Film erzählt weitere epische, mit Action vollgepackte Abenteuer des respektlosen Draufgängers Peter Quill alias Star-Lord, und seiner Bande exzentrischer Helden. Nachdem sie den Planeten Xandar und seine Bewohner gerettet und dadurch Popularität und Ruhm erlangt haben, fliegen die Guardians als Söldner durchs All, um das Universum zu beschützen.
